Optimization of keratin extraction from chicken feathers using Novo Pro-D® enzyme and Draper-Lin experimental design

Keratins are a family of structural proteins that can be isolated from a variety of animal tissues with protective functions, including chicken feathers. In the present study, the extraction percentage of keratin from chicken feathers (Gallus gallus) was optimized and subsequently characterized. Response Surface Methodology (RSM) was employed to optimize the extraction process, utilizing the Novo Pro-D® enzyme and a Draper-Lin experimental design. The extraction percentage (%E) served as the response factor, while independent variables included temperature (°C), extraction time (h), pH, and the enzyme-to-substrate (E/S) ratio. Feather samples were previously characterized through proximate analysis, determining moisture, ash, and crude fat contents using standard methods. Nitrogen (protein) content was determined via the Kjeldahl method. Proximate analysis revealed a protein content of 86.42%. The optimized variables were found to be: pH 6.0, temperature 30 °C, time 8 h, and an E/S ratio of 0.64, resulting in a 92% extraction yield. FTIR analysis confirmed characteristic protein signals associated with α-helix and β-sheet fractions, indicating successful keratin extraction under the applied conditions. The high extraction yield and favorable hydrolysis conditions demonstrate the feasibility of implementing a process parallel to the poultry industry, where these innovative techniques can enhance productivity and strengthen the regional sector.

Las queratinas son un grupo de proteínas estructurales que pueden obtenerse a partir de diversos tejidos animales con función protectora, como es el caso de las plumas de pollo. En este trabajo se optimizó el proceso de extracción de queratina presente en plumas de Gallus gallus y posteriormente se realizó su caracterización. Para optimizar la extracción se empleó la metodología de superficie de respuesta, utilizando la enzima Novo Pro-D® y un diseño experimental Draper-Lin. Como variable de respuesta se consideró el porcentaje de extracción (%E), mientras que las variables independientes fueron la temperatura, el tiempo de extracción, el pH y la relación enzima/sustrato (E/S). Previamente, las plumas fueron sometidas a un análisis proximal, en el que se determinaron humedad, cenizas y grasa cruda mediante métodos estándar, mientras que el contenido de nitrógeno (asociado a proteína) se evaluó por el método Kjeldahl. Los resultados evidenciaron un alto contenido proteico, alcanzando un valor de 86.42%. Las condiciones óptimas de extracción se establecieron en pH 6.0, temperatura de 30 °C, tiempo de 8 horas y una relación enzima/sustrato de 0.64, bajo las cuales se logró un rendimiento de extracción cercano al 92%. El análisis FTIR de la queratina obtenida confirmó la presencia de señales características de la proteína en su forma nativa, asociadas a estructuras de α-hélice y lámina β, lo que indica que, bajo las condiciones empleadas, fue posible extraer queratina conservando su integridad estructural. El alto rendimiento alcanzado, junto con las suaves condiciones de hidrólisis, evidencian el potencial de este proceso para ser implementado como una alternativa dentro de la industria avícola. Además, el uso de este tipo de estrategias innovadoras puede contribuir a mejorar la productividad y fortalecer el desarrollo industrial regional.
