El análisis de amplificación al azar de fragmentos polim6rficos de AON (RAPD) fue usado para caracterizar dos nuevas variedades de tabaco tipo Flue Cured y otra de tabaco negro derivadas por la técnica del cultivo de anteras in vitro. Los RAPO son propuestos como un complemento apropiado de las características morfoagronómicas para llenar los requisitos de los estándares internacionales para el registro de semillas, establecidos para la identificación de variedades de tabaco. La identificación de tres variedades de tabaco y sus progenitores fue realizada usando el análisis RAPO con 64 iniciadores, Un número de 214 productos polimórficos fueron amplificados desde 14 iniciadores.
El análisis estadístico realizado con el programa NTSYS, versión 1.2, usando el coeficiente de similitud de Jaccard. La inspección visual reveló que cinco iniciadores permitieron la separación de las variedades en dos grupos, de acuerdo con el tipo de tabaco: los Virginia (Flue Cured) y los Negros, mientras que un grupo de nueve iniciadores separó cada variedad y estableció las relaciones genéticas con sus progenitores. Los resultados obtenidos muestran que esta técnica es apropiada para establecer diferencias genéticas entre las variedades de tabaco.
Randomly Amplified Polymorphic DNA (RAPO) analysis was used to characterize two new Flue Cured and one black tobacco type varieties derived from in vitro anther tissue culture technique. RAPOs are proposed as an appropriate complement of the morphoagronomic characteristics evaluations to fulfil international seed registration standards established for the identification of tobacco varieties. The identification of three tobacco varieties and their parents was carried out using the RAPO analysis with 64 random primers. Polymorphic products, 214 in number, were amplified only from 14 primers. Statistical analysis realized with the NTSYS program version 1.2 using the Jaccard similarity coefficient. The visual inspection revealed that five primers allowed the separation of the varieties in two groups, according to the type of tobacco: the Flue Cured and Black; while a group of nine primers separates each variety and establish its genetic relationship with their parents. The results obtained show that this technique is appropiated to establish genetic differences between tobacco varieties.
