Reacción cuantitativa de la Rana Macho “Rana Pipiens” a la acción de Gonadotrofina Coriónica

En esta investigación se concluyo que la secreción de espermatozoides en ]a rana. macho Rana pipiens con la inyección de gonadotrofina coriónica es una reacción satisfactoria como procedimiento
de ensayo biológico para la determinación de la actividad de las preparaciones
de gonadotrofina coriónica. El procedimiento consistió en
inyectar soluciones de la hormona en el saco dorsal linfático de la
rana macho y dos horas más tarde observar en el microscopio la
presencia de espermatozoides en la orina.
Se construyeron curvas de frecuencia integral para dosis/reacción
para las reacciones de cada una de las cuatro horas de observación
después de la inyección.
Se encontró que dos horas después de la inyección es el periodo
de observación más adecuado.
EI promedio de la dosis de gonadotrofina requerida es 9.6 Unidades
Internacionales por rana.
In this investigation it was concluded that secretion of spermatozoa by
the male frog Raila pipiclls on injection of chorionic gonadotrophin is a suitable
reaction for a biological assay procedure for the determination of potency of
chorionic gonadotrophin preparations. The procedure consisted of injection 01
solutions of the hormone .into the dorsal lymph sac of the male frog, then after
two hours the urine is observed microscopically for spermatozoa.
Integrated frequency curves for dose/response were constructed for reactions
at hourly observation periods for four hours after the time of injection
and for frogs which had been used up to five times.
The most suitable for observing the secretion of spermatozoa was found to
1)(' two hours after injection. Frogs can be used four times with at least a seven
day interval betwen usc.
The average dose of gonadotrophin required is 9, 6 International Units per
frog.
