Effect of ibuprofen and paracetamol on the avoidance behavior and stability of the lysosomal membrane of the Eisenia fetida

En este estudio se evaluó la toxicidad de los fármacos antiinflamatorios no esteroideos, ibuprofeno y paracetamol, a través del bioensayo de comportamiento de evasión y tiempo de retención de rojo neutro (TRRN) para evaluar la estabilidad de la membrana lisosomal sobre la lombriz de tierra Eisenia fetida. La CE80 para la evasión fueron 54,8 mg/kg para ibuprofeno y 223,8 mg/kg para paracetamol. El TRRN, la concentración más baja a la cual se observa efecto (CBOE) de la pérdida de la estabilidad de la membrana lisosomal fue a los 5 mg/kg de ibuprofeno y 100 mg/kg de paracetamol. Estos resultados, indican que la presencia de ambos fármacos en suelos afecta la selección del hábitat y provocan daño celular en E. fetida. La prueba de tiempo de retención de rojo neutro fue más sensible que el de conducta de evasión y confirma que es un biomarcador de alerta temprana de efectos tóxicos.
This study evaluated the toxicity of the non-steroidal anti-inflammatory drugs ibuprofen and paracetamol in the earthworm Eisenia fetida using the avoidance behavior test and the neutral red retention time (NRRT) assay. The EC80 values for avoidance behavior were 54.8 mg kg⁻¹ for ibuprofen and 223.8 mg kg⁻¹ for paracetamol. The NRRT assay showed lysosomal membrane destabilization at concentrations as low as 5 mg kg⁻¹ for ibuprofen and 100 mg kg⁻¹ for paracetamol, corresponding to the lowest observed effect concentrations (LOEC). These results demonstrate that the presence of both pharmaceuticals in soil affects habitat selection and induces cellular damage in E. fetida. Moreover, the NRRTassay exhibited greater sensitivity than the avoidance behavior test, confirming its value as an early-warning biomarker of toxic effects.
